The RAB5 protein is a small GTPase involved in membrane trafficking in the early endocytic pathway. The protein encoded by this gene binds the GTP-bound form of the RAB5 protein preferentially over the GDP-bound form, and functions as a guanine nucleotide exchange factor for RAB5. The encoded protein is found primarily as a tetramer in the cytoplasm and does not bind other members of the RAB family. Mutations in this gene cause macrocephaly alopecia cutis laxa and scoliosis (MACS) syndrome, an elastic tissue disorder, as well as the related connective tissue disorder, RIN2 syndrome. Alternative splicing results in multiple transcript variants.

The protein encoded by this gene, known as RNase5 or angiogenin, is a member of the RNase A superfamily, though it has weak ribonucleolytic activity. It is a potent mediator of angiogenesis, binding to actin on endothelial cell surfaces to promote blood vessel formation. This protein also accumulates in the nucleolus to stimulate ribosomal transcription and, under stress, translocates to the cytosol to hydrolyze tRNAs, affecting protein synthesis. The mature protein, after cleavage of a signal peptide, contains a nuclear localization signal, a cell-binding motif, and a catalytic domain. It is neurotrophic, neuroprotective, and exhibits antimicrobial activity against bacteria and fungi, such as S. pneumoniae and C. albicans. This gene plays roles in cell growth and tumor progression, with mutations linked to amyotrophic lateral sclerosis (ALS). Alternative splicing produces multiple transcript variants encoding the same protein.
